The aging process of microplastics (MPs) could significantly change their physical and chemical characteristics and impact their migration behavior in soil. However, the complex effects of different cations and humic acids (HA) on the migration of aged MPs through saturated media are not clear. In this research, the migration and retention of pristine/aged PSMPs (polystyrene microplastics) under combined effects of cations (Na+, Ca2+) (ionic strength = 10 mM) and HA (0, 5, 15 mg/L) were investigated and analyzed in conjunction with the two-site kinetic retention model and DLVO theory. The findings showed that the aging process accelerated PSMPs migration under all tested conditions. Aged PSMPs were less susceptible to Ca2+ than pristine PSMPs. Under Ca2+ conditions, pristine/aged PSMPs showed higher retention than under Na+ conditions in the absence of HA. Furthermore, under Na+ conditions, the migration of aged PSMPs significantly increased at higher concentrations of HA. However, under Ca2+ conditions, the migration of aged PSMPs decreased significantly at higher concentrations of HA. In higher HA conditions, HA, Ca2+, and PSMPs interact to cause larger aggregations, resulting in the sedimentation of aged PSMPs. The DLVO calculations and two-site kinetic retention models' results showed the detention of PSMPs was irreversible under higher HA conditions (15 mg/L) with Ca2+, and aged PSMPs were more susceptible to clogging. These findings may help to understand the potential risk of migration behavior of PSMPs in the soil-groundwater environment.

The free radicals released from the advanced oxidation processes can enhance microplastics degradation, however, the existence of microbes acting synergistically in this process is still uncertain. In this study, magnetic biochar was used to initiate the advanced oxidation process in flooded soil. paddy soil was contaminated with polyethylene and polyvinyl chloride microplastics in a long-term incubation experiment, and subsequently subjected to bioremediation with biochar or magnetic biochar. After incubation, the total organic matter present in the samples containing polyvinyl chloride or polyethylene, and treated with magnetic biochar, significantly increased compared to the control. In the same samples there was an accumulation of "UVA humic" and "protein/phenol-like" substances. The integrated metagenomic investigation revealed that the relative abundance of some key genes involved in fatty acids degradation and in dehalogenation changed in different treatments. Results from genome-centric investigation suggest that a Nocardioides species can cooperate with magnetic biochar in the degradation of microplastics. In addition, a species assigned to the Rhizobium taxon was identified as a candidate in the dehalogenation and in the benzoate metabolism. Overall, our results suggest that cooperation between magnetic biochar and some microbial species involved in microplastic degradation is relevant in determining the fate of microplastics in soil.

Considering the potential effect of the ambient temperature on soil microorganisms during heavy metal immobilization by hydrochar, 60 days of soil incubation was conducted to explore the impact of ambient temperature (5, 25, and 35 °C) on the immobilization of Pb and Zn by chitosan-magnetic sawdust hydrochar (CMSH) and magnetic chitosan hydrochar (MCH). The results showed that soil pH was relatively high and total organic carbon (TOC) was slightly lower in the 35 °C treatment. The diethylenetriaminepentaacetic acid (DTPA) available state content decreased significantly with the temperature increasing. Meanwhile, the ratios of stable Pb and Zn in the sequential extraction method proposed by the European Community Bureau of Reference (BCR) gradually increased with increasing temperature. The heatmap based on microbial community showed that elevated temperature not only favored the enrichment of metal-stable phyla, such as Chloroflexi, but was also involved in inhibiting the growth of Firmicutes, Actinobacteriota, and Proteobacteria. Meanwhile, different genera (Fonticella and Bacillus) in the Firmicutes phylum had distinct responses to temperature as well as to heavy metal immobilization effects. Subsequently, redundancy analysis confirmed that Chloroflexi and Fonticella were positively correlated with temperature and stable state metal content, while Actinobacteriota and Bacillus were negatively correlated with temperature and were positively correlated with DTPA available metal content. Moreover, Pb and Zn indicators displayed significant correlations for the dominant genera (R2 > 0.8, p < 0.02).

Microplastics (MPs) are susceptible to aging in the environment, and aged MPs are highly migratory in soil due to their smaller particle size and more negative surface charge, but the effects of soil environmental factors on the fate and transport of aged MPs are still unclear. In this study, the transport behavior of pristine/aged MPs in unsaturated sandy porous media was examined under different ionic strength (IS), cationic type (Na+, Ca2+) and humic acid (HA) conditions. The results indicated that the surface charge, surface oxygen-containing functional groups and surface morphology of MPs changed significantly after aging, and that the mobility of aged MPs was significantly enhanced than the pristine MPs under all test conditions. The retention amounts of pristine/aged MPs in unsaturated porous media increased with IS, and IS had a less inhibitory effect on the transport of aged MPs than pristine MPs. The mobility of pristine/aged MPs in Ca2+ solutions was significantly weaker than that in Na+ solutions due to enhanced straining and electrostatic adsorption. HA promoted the mobility of pristine/aged MPs in unsaturated porous media under all IS Na+ (1, 10, and 25 mM) solutions and lower IS (1 mM) Ca2+ solutions, and the ability of HA to promote the transport of aged MPs was significantly stronger than that of pristine MPs due to the higher adsorption of HA on the surface of aged MPs. However, at higher IS (10 mM) Ca2+ solution conditions, the bridging effect of Ca2+ led to the formation of HA-MPs complexes, which altered the hydrophobicity of the pristine/aged MPs surface and the pristine/aged MPs were mainly retained on the air-water interface (AWI). CFT theory and two-site kinetic retention models indicated that the retention of pristine/aged MPs in unsaturated media was dominated by monolayer adsorption, straining and clogging effects. The current research findings may provide insights into the fate and transport of aged MPs in soil and their potential risk of groundwater contamination.

Magnetically modified biochar, with a rougher surface and more positive surface charge, may interact with microplastics (MPs) after being applied to soil, potentially altering the fate and transport of MPs in porous media. In this study, the transport and retention behavior of polystyrene microplastics (PSMPs) in a sandy porous media mixed with biochar/Fe3O4 modified biochar (Fe3O4-biochar) was investigated under various chemical conditions (humic acid (HA), ionic strength (IS) and cationic types (Na+/Ca2+)). The results showed that the addition of biochar and Fe3O4-biochar can hinder the transport of PSMPs in porous media without HA, and that Fe3O4-biochar was more effective in inhibiting the transport of PSMPs through electrostatic adsorption and complexation, with an optimum retention efficiency of 92.36 %. HA significantly attenuated the retention of PSMPs in both porous media through electrostatic repulsion, steric resistance and competitive adsorption under 1 mM Na+ solutions, and the mobility of PSMPs in Fe3O4-biochar/sand was enhanced more significantly than in biochar/sand with the increase of HA concentration. IS significantly inhibited the transport of PSMPs in both porous media in the absence of HA, but there was an antagonistic effect of HA and IS on the transport of PSMPs in the presence of HA, with the facilitative effect of HA being stronger than the inhibitory effect of IS. Ca2+ was consistently more effective in inhibiting the transport of PSMPs than Na+ under all test conditions, and HA promoted the transport of PSMPs in all Na+ solutions, while it inhibited the transport of PSMPs in high IS (10 mM) with Ca2+ solutions. In addition, HA, Fe3O4-biochar and PSMPs tend to form larger aggregates under the complex interactions of Ca2+, leading to increased retention of PSMPs in porous media. The two-site kinetic retention models suggested that the retention of PSMPs in porous media with biochar was predominantly reversible attachment effect, while retention in porous media with Fe3O4-biochar was predominantly an irreversible straining effect.

A series of 60-day soil immobilized incubations were performed to explore the impacts of various factors (incubation time, chitosan modified magnetic sawdust hydrochar (CMSH) dosages, initial pH values, moisture contents, and humic acid (HA)) on CMSH immobilization of Pb and Zn. DTPA and BCR extraction techniques were undertaken to study the distribution of form transformations of Pb and Zn. CMSH showed significant immobilization ability for both DTPA-Pb and DTPA-Zn, and the highest removal rates were shown to be 57.40% and 90.00% for Pb and Zn respectively. After 60 days of incubation, the residual Pb was enhanced by 34-61% and residual Zn increased by 25-41%, which indicated that CMSH was effective in immobilizing Pb and Zn. Meanwhile, the immobilization efficiency improved with increasing incubation time, CMSH dosage, HA dosage, and initial solution pH. In particular, 5% HA application increased the soil TOC and accelerated the metal stabilization processes, with the residual forms of Pb and Zn eventually reaching a maximum of 73% and 71%, respectively. In addition, the alkaline initial solution promoted the ion exchange, surface complexation reaction, and cationic-π interaction, resulting in a better immobilization of Pb and Zn by CMSH. Finally, according to the orthogonal analysis of BCR results, HA dosage was the major factor affecting Pb and Zn immobilization by CMSH compared to soil pH and moisture content in this study.

Biochar and hydrochar, as valuable and eco-friendly soil remediation materials from greenwaste, have potential to enhance methane oxidation in paddy soil. But the mechanism of biomass carbon on the improvement of methane-oxidizing bacteria communities in paddy soil has not been adequately elucidated. In the present study, the effect of different-temperature rice straw-based biomass carbon (RB400, RB600, RH250 and RH300) on methane oxidation were investigated by analyzing the soil dissolved organic matter (DOM), physicochemical properties and changes in microbial community structure. The results of the 17-day incubation experiment showed that the methane oxidation rate increased under all types of biomass carbon in the first 6 days. The enhancement of methane oxidation rate was more pronounced for biochar compared to hydrochar, with RB600 being the most effective treatment. The result of excitation-emission matrix (EEM) fluorescence spectroscopy showed that less DOM were released from the soil in the biochar treatments compared to the hydrochar treatments and protein-like were detected only in the hydrochar group. Microbial analysis further showed that hydrochar inhibited the growth of Bacillus, Methylobacter, and Methylocystis, while RB600 significantly increased the relative abundance of methanotrophs (responsible for methane oxidation), such as Methylocystis and Methylobacter, which was consistent with their different effects on the methane oxidation rate. Moreover, from the analysis of principal component analysis (PCA) and canonical correspondence analysis (CCA), Methylobacter and Methylocystis were negatively respond to H/C of biomass carbon. The present study provides a deeper understanding of the effect of biomass carbon obtained by different processes on methane oxidation when applied to soil from the perspective of organic matter and microbial communities.

The preparation of magnetic biochar from sewage sludge and rice straw for heavy metal contaminated soil remediation has greater application prospects, but its remediation mechanism was rarely considered by combining soil physicochemical properties with microbial community. In this study, the effects of magnetic sewage sludge biochar (SSB) and rice straw biochar (RSB) on Cd and Pb immobilization in paddy soil were compared and analyzed by 60-day soil incubation experiments. The results illustrated that DTPA-Cd and DTPA-Pb were reduced by 51.53% (43.07%) and 53.57% (50.47%), while the percentage of residual fraction of the BCR procedure was enhanced by 31.27% (30.78%) of Cd and 27.25% (23.22%) of Pb in the SSB (RSB) treatment, respectively. Fe was detected on both SSB and RSB surfaces, but SSB had rougher and a larger specific surface area compared to RSB. The addition of SSB and RSB in paddy soil increased soil pH and TOC content, and affected the diversity and species of soil microbial community. Compared with the CK group, the relative abundance of Proteobacteria, Bacteroidota, and Lysobacter decreased, and the relative abundance of Actinobacteriota, Pontibacter, and Alkaliphilus increased with SSB and RSB treatments, all of which reflected the bioavailability of Cd and Pb reduction.

Biochar particles are extensively used in soil remediation and interact with microplastics (MPs), especially metal oxide-modified biochar may have stronger interactions with MPs. The mechanism of interactions between humic acid (HA) and different valence cations is different and the co-effect on the transport of MPs is not clear. In this study, the co-effects of HA and cations (Na+, Ca2+) on the transport and retention of MPs in saturated porous media with peanut shell biochar (PSB) and MgO-modified PSB (MgO-PSB) were systematically investigated. Breakthrough curves (BTCs) of MPs were fitted by the two-site kinetic retention model for analysis. In the absence of HA, the addition of PSB and MgO-PSB significantly hindered the transport of MPs in saturated porous media, and the retention of MPs increased from 34.2% to 59.1% and 75.5%, respectively. In Na+ solutions, the HA concentration played a dominant role in controlling MPs transport, compared to the minor role of Na+. The transport capacity of MPs always increased gradually with the increase of HA concentration. Whereas, in Ca2+ solutions, Ca2+ concentrations had a stronger effect than HA. The transport ability of MPs was instead greater than that in Na+ solutions as the HA concentration increased at low ionic strength (1 mM). However, the transport capacity of MPs was significantly reduced with increasing HA concentrations at higher ionic strength (10, 100 mM). The two-site kinetic retention model indicated that chemical attachment and physical straining are the main mechanisms of MPs retention in the saturated porous media.

As a promising amendment, biochar has excellent characteristics and can be used as a remediation agent for diverse types of soil pollution. Biochar is mostly made from agricultural wastes, forestry wastes, and biosolids (eg, sewage sludge), but not all the biochar has the same performance in the improvement of soil quality. There is a lack of guidelines devoted to the selection of biochar to be used for different types of soil pollution, and this can undermine the remediation efficiency. To shed light on this sensitive issue, this review focus on the following aspects, (i) how feedstocks affect biochar properties, (ii) the effects of biochar on heavy metals and organic pollutants in soil, and (iii) the impact on greenhouse gas emissions from soil. Generally, the biochars produced from crop residue and woody biomass which are composed of lignin, cellulose, and hemicellulose are more suitable for organic pollution remediation and greenhouse gas emission reduction, while biochar with high ash content are more suitable for cationic organic pollutant and heavy metal pollution (manure and sludge, etc.). Additionally, the effect of biochar on soil microorganisms shows that gram-negative bacteria in soil tend to use WB biochar with high lignin content, while biochar from OW (rich in P, K, Mg, and other nutrients) is more able to promote enzyme activity. Finally, our recommendations on feedstocks selection are presented in the form of a flow diagram, which is precisely intended to be used as a support for decisions on the crucial proportioning conditions to be selected for the preparation of biochar having specific properties and to maximize its efficiency in pollution control.

Heavy metals in the soil are major global environmental problems. Waste cotton was used to synthesize a novel ß-CD/hydrothermal biochar (KCB), which is a low-cost and environment-friendly adsorbent for heavy metal soil remediation. KCB were used as reactive materials of electrokinetic-permeable reactive barrier (EK-PRB) to explore the removal characteristics of heavy metals. FTIR and XPS analysis revealed that KCB contained large numbers of surface functional groups. Adsorption of KCB for Pb2+ and Cd2+ reached 50.44 mg g-1 and 33.77 mg g-1, respectively. Metal ions in contaminated soil were removed by reactive barrier through electromigration, electrodialysis and electrophoresis, the removal efficiency of Pb2+ and Cd2+ in soil reached 92.87% and 86.19%. This finding proves that KCB/EK-PRB can be used as a cheap and green process to effectively remediate soils contaminated with heavy metals.

The application of phenylurea herbicides (PUHs) may lead to the extensive distribution in soils, while the role of straw biochar as a soil amendment on the transport and sorption of PUHs are still unclear. Thus, the transport and sorption behavior of three typical PUHs with rice straw biochar (RSB) was studied in both adsorption simulation experiments of aqueous solution and packed column experiments. The sorption mechanism of RSB to herbicides was investigated through batch sorption studies with three influencing factors including dosage of RSB, pH, and ionic strength (IS) with orthogonal test. The sorption coefficients were improved significantly by increasing the dosage of RSB, while there was no obvious influence by enhancing the pH and IS value. The optimal sorption conditions (pH value at 3, IS at 0.1 M, and RSB dosage at 60 mg) of three herbicides were set and the maximum removal rates of Monuron, Diuron, and Linuron were 41.9%, 25%, and 56.8%, respectively. The co-transport process of RSB and PUHs were investigated under different RSB dosage, pH value, and IS value. The retention effect increased greatly with enhancing the RSB dosage and pH value. However, IS did not have a significant influence on the retention of RSB, and therefore it had little effect on the adsorption capacity, which was consistent with the results of sorption experiments. The breakthrough curves (BTCs) for co-transport were well simulated by the two-site non-equilibrium convection-dispersion equation (CDE). Most of the regression coefficients (R2) were above 0.99, which uncovered the co-transport in packed column were affected by physical absorption and chemical forces. According to the fitting parameters analysis, the RSB particles and PUHs were subjected to a greater resistance and a stronger stability by reducing pH value in porous media. The presence of RSB increased the amount of dynamic sorption sites in the entire co-transport system, which led to a significant promotion of the PUHs' sorption and interception.

Biochar can be used as a soil amendment. However, it generally possesses unique physicochemical properties and complex organics, which could affect soil methanogenesis. In this study,straw-based biochars obtained at 300 °C (BC300), 500 °C (BC500) and 700 °C (BC700) were added to the paddy soil. Compared with the blank group, BC300 significantly increased paddy soil methane emissions by about 38%. However, this promoting effect gradually disappeared with the increase of pyrolysis temperature, and the inhibition even appeared in the BC700 group with the methane reduction by 18.2%. This might be related to the organics released from biochar. Van Krevelen (VK) diagram showed that the aromaticity of BC700 and BC500 were significantly higher than BC300. Fluorescent analysis further revealed that BC300 increased the amount of degradable fluorescent organics in the soil, which could provide more substrate for methane production. Moreover, as pyrolysis temperature increased, the fluorescent organics released were more likely to be non-biodegradable humus. In addition, it was shown that BC700 could adsorb some inherent organics in the soil, and thus reduced the total organic content and inhibited soil methane emissions. Microbial analysis showed that methanogenesis had a positive correlation with the abundance of syntrophic bacteria (e.g. Desulfobacca and Clostridium) which had ability to further degrade various types of organics and provided substrates to the methanogens. This article provides a deeper understanding regarding for the effects of biochar on methane emission from paddy soil in terms of organics and microbial perspectives.

Directly returning rice straw to the paddy soil would significantly stimulate methane emission, and hydrochar has potential to be used as soil conditioner. However, the effects of hydrochar on the methane emission from paddy soil and the related mechanisms are still unclear. In the present study, straw-based hydrochar obtained at 200 °C (HC200), 250 °C (HC250) and 300 °C (HC300) and hydrochar after removal of bio-oil at these temperatures (CHC200, CHC250, and CHC300) were prepared and added to the paddy soil. The application of HC200, HC250 and HC300 resulted in the enhanced methane production compared to the control, showing 4.3, 1.6 and 1.5-fold higher methane production, respectively. It was related to the large amount of dissolved organic matter (DOM) released from hydrochar. Excitation-emission matrix fluorescence spectroscopy with parallel factor analysis (EEM-PARAFAC) showed that the hydrochar-derived DOM mainly included humic-like, phenolic and less aromatic structures, and with the increase of hydrothermal temperature, the content of humic-like substances and phenols increased, while biodegradable organics decreased. This was consistent with the maximum methane production by HC200. After incubation, there was no low-aromatic structures observed in the soil leachate, and the residual organics were mainly humus. The EEM-PARAFAC results were supported by compositional characterization of soil leachate by high-resolution mass spectrometry, and the refractory organics released from hydrochar was mainly lignins or (CRAM)-like structures in the range of H/C = 0.8-1.6 and O/C = 0.1-0.5. The organics dissolved from the washed hydrochar was significantly reduced, and some washed hydrochar (CHC250 and CHC300) even inhibited methane emission possibly due to their ability to adsorb organics. Microbial analysis further showed that the increased methane production resulted from hydrochar was associated with the enrichment of Janibacter, Anaeromyxobacter, Anaerolinea and Sporacetigenium. This present study provided a better understanding to the effect of hydrochar on methanogenesis in paddy soil.

Hydraulic fracturing is expanding rapidly in the US to meet increasing energy demand and requires high volumes of hydrofracking fluid to displace natural gas from shale. Accidental spills and deliberate land application of hydrofracking fluids, which return to the surface during hydrofracking, are common causes of environmental contamination. Since the chemistry of hydrofracking fluids favors transport of colloids and mineral particles through rock cracks, it may also facilitate transport of in situ colloids and associated pollutants in unsaturated soils. We investigated this by subsequently injecting deionized water and flowback fluid at increasing flow rates into unsaturated sand columns containing colloids. Colloid retention and mobilization was measured in the column effluent and visualized in situ with bright field microscopy. While <5% of initial colloids were released by flushing with deionized water, 32-36% were released by flushing with flowback fluid in two distinct breakthrough peaks. These peaks resulted from 1) surface tension reduction and steric repulsion and 2) slow kinetic disaggregation of colloid flocs. Increasing the flow rate of the flowback fluid mobilized an additional 36% of colloids, due to the expansion of water filled pore space. This study suggests that hydrofracking fluid may also indirectly contaminate groundwater by remobilizing existing colloidal pollutants.

The prediction of colloid transport in unsaturated porous media in the presence of large energy barrier is hampered by scant information of the proportional retention by straining and attractive interactions at surface energy minima. This study aims to fill this gap by performing saturated and unsaturated column experiments in which colloid pulses were added at various ionic strengths (ISs) from 0.1 to 50 mM. Subsequent flushing with deionized water released colloids held at the secondary minimum. Next, destruction of the column freed colloids held by straining. Colloids not recovered at the end of the experiment were quantified as retained at the primary minimum. Results showed that net colloid retention increased with IS and was independent of saturation degree under identical IS and Darcian velocity. Attachment rates were greater in unsaturated columns, despite an over 3-fold increase in pore water velocity relative to saturated columns, because additional retention at the readily available air-associated interfaces (e.g., the air-water-solid [AWS] interfaces) is highly efficient. Complementary visual data showed heavy retention at the AWS interfaces. Retention by secondary minima ranged between 8% and 46% as IS increased, and was greater for saturated conditions. Straining accounted for an average of 57% of the retained colloids with insignificant differences among the treatments. Finally, retention by primary minima ranged between 14% and 35% with increasing IS, and was greater for unsaturated conditions due to capillary pinning.

This study demonstrates that the pattern assembly and attachment strength of colloids in an evaporating sessile droplet resting on a smooth substrate can be controlled by adding nonionic solutes (surfactant) to the solution. As expected, increasing the surfactant concentration leads to a decrease in initial surface tension of the drop, σ(0). For the range of initial surface tensions investigated (39-72 mN m(-1)), three distinct deposition patterns were produced: amorphous stains (σ(0) = 63-72 mN m(-1)), coffee-ring stains (σ(0) = 48-53 mN m(-1)), and concentric rings (σ(0) = 39-45 mN m(-1)). A flow-displacement system was used to measure the attachment strength of the dried colloids. Characteristic drying regimes associated with the three unique pattern formations are attributed to abrupt transitions of contact line dynamics during evaporation. The first transition from slipping- to pinned-contact line was found to be a direct result of the competition between mechanical instability of the droplet and the friction generated by pinned colloids at the contact line. The second transition from pinned- to recurrent-stick-rip-slip-contact line was caused by repeated liquid film rupturing from evaporation-intensified surfactant concentration. Data from flow-displacement tests indicate that attachment strength of dried particles is strongest for amorphous stains (lowest surfactant concentration) and weakest for concentric rings (highest surfactant concentration). The mechanism behind these observations was ascribed to the formation and adsorption of micelles onto colloid and substrate surfaces as the droplet solution evaporates. The range of attachment forces observed between the colloids and the solid substrate were well captured by extended-DLVO interactions accounting for van der Waals attraction, electric double layer repulsion, and micelle-protrusion repulsion. Both empirical and theoretical results suggest that an increasingly dense layer of adsorbed micellar-protrusions on colloid and substrate surfaces acts as a physical barrier that hinders strong van der Waals attractive interactions at close proximity. Thereby, colloid stains dried at higher surfactant concentrations are more easily detached from the substrate when dislodging forces are applied than stains dried at lower surfactant concentrations.

This study examined the impacts of bacteria on the algal biomass, lipid content and efficiency of wastewater treatment during the heterotrophic cultivation of Chlorella pyrenoidosa. Our results showed that soybean-processing wastewater can enhance the accumulation of lipids in algal cells and thus raise the lipid yield in the pure culture. The bacteria coexisting with algae improved the degradation of total nitrogen (TN), total phosphorus (TP), glucose and chemical oxygen demand (COD). Although the biomass productivity of algae was not significantly affected, the total algal lipid content and lipid production rate were slightly reduced when bacteria coexisted with algae. The difference in the compositions of the medium is presumed to be the main contributing factor for the variation in total lipid content in presence and absence of bacteria. The TN, TP, and COD decreased during the assimilatory process undertaken by C. pyrenoidosa, and the removal efficiency of TN by bacteria depended on the type of nitrogen species in the medium. Additionally, the apparent interaction between the bacterial and algal cultures varied with the changes in experimental conditions. Algae could compete with bacteria for the carbon and energy sources, and inhibit the growth of the bacteria in the presence of high organic matter concentration in the medium.

As one kind of emerging contaminant, synthetic musks (SMs) are commonly used in varying amounts in many personal care products and have been detected in different environmental systems. Occurrence and fate of four common SMs [galaxolide (HHCB), tonalide (AHTN), musk xylene (MX) and musk ketone (MK)] were studied in different sewage treatment plants (STPs) of Shanghai, China among different seasons. Results showed that total dissolved concentrations of the four SMs were 536-3,173 ng/L in influent, 351-2,595 ng/L in effluent and 147-6,839 µg/kg dry weights in sludge. The SM concentrations varied with input sources, STP treatment processes, usage patterns, and different seasons or surveyed years of consumption. There was no significant removal of SMs in most of the sewage samples of the four STPs. Significant positive correlations were observed between concentrations of HHCB and AHTN (R(2) = 0.9062, n = 12, p< 0.05), HHCB and MK (R(2) = 0.7471, n = 8, p< 0.05), as well as AHTN and MK (R(2) = 0.9321, n = 8, p< 0.05) in sludge samples.

Porcine reproductive and respiratory syndrome virus (PRRSV) can subvert early innate immunity, which leads to ineffective antimicrobial responses. Overcoming immune subversion is critical for developing vaccines and other measures to control this devastating swine virus. The overall goal of this work was to enhance innate and adaptive immunity following vaccination through the expression of interferon (IFN) genes by the PRRSV genome. We have constructed a series of recombinant PRRS viruses using an infectious PRRSV cDNA clone (pCMV-P129). Coding regions of exogenous genes, which included Renilla luciferase (Rluc), green and red fluorescent proteins (GFP and DsRed, respectively) and several interferons (IFNs), were constructed and expressed through a unique subgenomic mRNA placed between ORF1b and ORF2 of the PRRSV infectious clone. The constructs, which expressed Rluc, GFP, DsRed, efficiently produced progeny viruses and mimicked the parental virus in both MARC-145 cells and porcine macrophages. In contrast, replication of IFN-expressing viruses was attenuated, similar to the level of replication observed after the addition of exogenous IFN. Furthermore, the IFN expressing viruses inhibited the replication of a second PRRS virus co-transfected or co-infected. Inhibition by the different IFN subtypes corresponded to their anti-PRRSV activity, i.e., IFNω5 ° IFNα1 > IFN-ß > IFNδ3. In summary, the indicator-expressing viruses provided an efficient means for real-time monitoring of viral replication thus allowing high­throughput elucidation of the role of host factors in PRRSV infection. This was shown when they were used to clearly demonstrate the involvement of tumor susceptibility gene 101 (TSG101) in the early stage of PRRSV infection. In addition, replication­competent IFN-expressing viruses may be good candidates for development of modified live virus (MLV) vaccines, which are capable of reversing subverted innate immune responses and may induce more effective adaptive immunity against PRRSV infection.
